Next generation sequencing by the method of Roche/454 is available at Yale University. The cost is subsidized by the university, and charges to you are generally only for recouping our costs. Charges are usually made to your grant VIP number.
The quantities required for different library sequencing projects are:
Genomic DNA 5 µg
cDNA 1 µg
Amplicon 0.5µg
You bring the appropriate quantity of DNA to us, we do the rest. DNA sequence results are in FASTA format and will be sent to you via ftp.
Typical results are:
400,000 reads (clones)
100M bases of total sequence
240nt length
Prices for Roche/454 DNA sequencing:
A. Preliminary sequences (Titration, done to quantitate optimal ratio of DNA to bead; but results in a few thousand reads averaging 110 bases.) $3,051.12
B. Full 454 Sequencing Prep $5,802.24
Total cost for one reaction of a single library (Titration + Full sequence)
$8,853.36
